05Half-life, peak time, and the 36-hour study window
Mean terminal half-life in healthy adults is 17.5 hours. Oral clearance is about 2.5 L/h. Peak after a swallowed tablet sits near two hours. Food does not move Cmax or AUC in a clinically meaningful way, which is why the label says take it with or without meals.
A timed intercourse study put men on placebo, 10 mg, or 20 mg and assigned attempts at 24 and 36 hours. Rifampin-type inducers can flatten the window you paid for.

Hemodialysis between 24 and 30 hours after a dose contributed little to tadalafil or metabolite removal. 08When a different PDE5 or regimen fits better
Need a short clock and a meal you can actually time? Sildenafil 25-50 mg covers the usual starting strengths. Need a fast peak and you have already cleared QT drugs? Vardenafil starts at 10 mg, ceiling 20 mg. Need untimed Tuesdays as well as Fridays? Daily tadalafil 2.5 or 5 mg, not a 10 mg split.

Headache and dyspepsia remain the more familiar first-night complaints. They usually fade in hours. The delayed lumbar ache is the one that makes men think they caught something on Saturday.
